Mechanism:

  • G= increase in circulating Insulin-like Growth Factor (IGF) levels [R, R2]. AG  individuals had a 5.6 ng/ml increase from AA individuals, while those with the GG genotype had an 11.1 ng/ml increase[R].

The "G" allele is associated with:

  • Higher breast density and dense area with each G allele, based on studies of  Dutch [R] and caucasian Canadian [R]  premenopausal women. Higher breast density has been linked to higher risk of breast cancer [R].

  • Slightly higher risk of breast cancer [R].

  • Reduced risk of glioma (brain or spinal tumor) [R].

  • Longer overall survival in Metastatic Colorectal Cancer patients treated with the drug Bevacizumab [R].

  • Poorer prostate cancer survival. This is based on a long term study of  Japanese men with prostate cancer with bone metastasis [R].

The "A" allele is associated with:

  • Shorter overall survival in Metastatic Colorectal Cancer patients treated with the drug Bevacizumab, who have the AA genotype [R].
